Ushigome-yanagichō Station opened on 12 December 2000 as a Toei Ōedo Line stop in Haramachi 2-chōme, Shinjuku, on the route inherited from the former Toden tram service 13 between Shinjuku and Suitengūmae that closed in 1970. Although the working name in planning had been simply Yanagichō, the final name retained the historic Ichigaya-Yanagichō neighbourhood from the old Ushigome ward to distinguish it from other Yanagichō sites; the underground island platform sits beneath the Ichigaya-Yanagichō crossing of Ōkubo-dōri and Gaien-Higashi-dōri. PASMO acceptance began on 18 March 2007, and a new south-east exit with elevator entered service on 9 September 2017.

Toei's opening forecast put daily ridership at about 7,000 passengers; by fiscal 2024 the station was averaging 19,954 daily entries and exits — almost three times the original projection.
