Uguisudani Station opened on 11 July 1912 as a Railway Board stop on what was already classed as the Tōhoku Main Line, but because platforms were laid only on the loop-style Yamanote Line tracks the station effectively began life as a Yamanote stop. On 1 November 1925 the Keihin Line was extended to Tabata along the same metals, adding a second operating route through the station. The Yamanote and Keihin-Tōhoku tracks were quad-tracked on 19 November 1956 and the station gained a second island platform. JR East took over at JNR's privatisation on 1 April 1987, the Keihin-Tōhoku Line's daytime rapid service introduced on 13 March 1988 has bypassed the station ever since, Suica was accepted from 18 November 2001, and smart platform-edge doors entered service on the Keihin-Tōhoku platforms on 27 April 2024.

Honouring the bush-warbler (uguisu) in its name, the station broadcasts recorded warbler song from platform speakers during morning hours.
