Tsukigaseguchi Station opened on 28 December 1951 as a new general (passenger and freight) station of Japanese National Railways on the Kansai Main Line, inserted between Shimagahara and Ōkawara. Freight operations ended on 1 February 1962, and the station was destaffed on 1 May 1983 when luggage handling was abolished. With the privatisation of JNR on 1 April 1987 it passed to JR West. ICOCA IC-card service began on 13 March 2021, and the Kameyama Rail Operations Office was abolished on 1 July of the same year. The station building was rebuilt in December 1965.

Although the station is administratively in Minamiyamashiro, Kyoto Prefecture, its name reflects its role as the gateway to the Tsukigase plum-blossom district of Nara Prefecture's Nara City. It is also the easternmost station in Kyoto Prefecture, and its platform displays a large pillar advertising the Tsukigase Plum Grove.
