Kasama Station opened on 16 January 1889 as a station of the original Mito Railway. Ownership transferred to Nippon Railway on 1 March 1892, the line was nationalised on 31 October 1906, and the route was named the Mito Line in 1909. From 22 November 1915 the Kasama Hand-Push Tramway (later Kasama Inari Tramway) operated between the station forecourt and Kasama town, ceasing in 1930. The original wooden depot remains in use. JR East assumed control at privatisation on 1 April 1987, Suica service began on 18 November 2001, and the footbridge was rebuilt with an elevator in 2006.

Kasama's departure melodies feature songs by Kyu Sakamoto: Track 1 plays "Sachiura nara Te o Tatakō" ("If You're Happy and You Know It") and tracks 2 and 3 use "Letkiss," reflecting the singer's Kasama family ties.
