Kamimachi Station opened on 1 May 1925 as a stop on the Tamagawa Electric Railway. The Setagaya Line, on which the station now sits, was split off as an independent operation when the parent Tamagawa Line was abolished in 1969, and is today operated by Tōkyū. Station numbering was introduced in 2007, assigning Kamimachi SG06.

Kamimachi has a distinctive zigzag (chidori-shiki) layout in which the two side platforms sit diagonally on either side of a level crossing, separated by the depot. Each platform is four cars long, normally fenced in the middle; when rolling-stock exchanges take place, the fences are opened and two two-car sets stand end-to-end so passengers can switch trains. The down-side platform once had a Tamagawa-side shop and ticket office, but the office closed in April 2022 and only a vending machine remains; the up-side platform houses the only restroom on the entire Setagaya Line.
